Can a Lung Cancer Cell Scratch Assay Establish an Effect on Metastasis?

What the experiment observed

Milian and colleagues studied THC and CBD in three non-small-cell lung cancer cell lines. In one assay, they scratched a gap in a confluent cell layer and measured how much of that gap was recolonized under selected conditions. The authors reported that the cannabinoids reduced the measured in vitro migration endpoint. They also ran separate assays for proliferation and markers of cell phenotype.

Why a gap is a limited model

Gap closure is a measurable event in a controlled culture. It depends on how cells behave in that system. Metastasis in a person also involves invasion, survival during transport and growth at a distant site, along with interactions with surrounding tissues. Those steps are outside the scratch assay. The study’s tumor receptor-expression analysis was separate from the drug treatment of cultured cells.

How to assess a claim based on this study

Ask which endpoint the cited figure actually measures, what concentrations were used and whether the inference involves movement in a dish or an outcome in patients. The cell data can inform a mechanism hypothesis. Evidence that a product changes the spread of cancer in people would require a design that directly evaluates that outcome.
